Maturasi afinitas atau pematangan afinitas adalah proses peningkatan afinitas limfosit B dalam mengikat antigen.

Limfosit B, sebagai salah satu komponen imunitas, bekerja dengan mengeluarkan antibodi untuk melawan bakteri, virus, jamur, atau "penyusup" lain yang masuk ke dalam tubuh. Untuk dapat menghancurkan para penyusup ini, sel-sel imun harus mengenali mereka. Pengenalan terjadi saat antibodi berikatan dengan antigen yang ada di permukaan sel bakteri, virus, jamur, atau sel kanker.
