normalisation of the data by area Explanation: As another suggestion, and a bit of explanation: Most multi-variate analysis techniques (of which principal component analysis, or PCA is one) work best when the data set is scaled in some way (e.g. so the values for each variable lie between zero and one) and centred (e.g. by subtracting the mean, so the average value is zero). The scaling is often done by normalising to the peak area or height (e.g. dividing by the area of the biggest peak). This helps avoid peaks with high sensitivity factors (and thus intensities) dominating the analysis. For example, if peak A has values between 30,000 and 50,000 counts, and peak B has values between 0 and 5,000 counts, the large absolute values of peak A appear to describe most of the data, while peak B is actually showing a wider range of variation. By normalising to the maximum peak area, peak A and peak B have similar weight in the analysis. |
