Le réfractomètre compare en sortie de colonne l’indice de réfraction de la fraction éluée à celui du solvant pur selon l’équation :

n_(solvant polymère) représente l’indice de réfraction du mélange solvant-polymère
n_(solvant) est l’indice de réfraction du solvant
C est la concentration en polymère de la fraction éluée traversant le détecteur à l’instant t
et dn/dC est l’incrément d’indice de réfraction. Il est caractéristique d’un couple solvant-polymère.
La déviation du faisceau incident est donc dépendant de la concentration en polymère de la fraction éluée traversant le détecteur à l’instant t. Le réfractomètre est un détecteur de concentration, son signal est proportionnel à C (en masse.volume-1).

OPERATING PRINCIPLE OF THE REFRACTIVE INDEX DETECTOR
The refractive index (RI) detector compares the refractive index of the eluted fraction as it leaves the column to that of the pure solvent, using the following equation:
n_(solvent polymer) = n_(solvent) ˖ C×dn/dC
Where
n_(solvent polymer) is the refractive index of the solvent-polymer mixture
n_(solvent) is the solvent’s refractive index
C is the polymer concentration of the eluate as it goes through the detector
and dn/dC is the refractive index increment, a value specific to each solvent-polymer pair.
By how much the incident light beam deviates therefore depends on the polymer concentration of the eluate going through the detector at that time. The refractive index detector is concentration-dependant; its response signal is proportional to C (expressed as weight.volume-1).

Thermogravimetric analysis was used to study the sublimation and vaporisation processes of S(-) efaroxan hydrochloride, a potent and highly selective α-2-adrenoreceptor antagonist. The kinetic parameters of the sublimation were investigated by conventional, isothermal thermogravimetric analysis (TGA) and modulated TGA (MTGA). The physico-chemical characterisation of the sublimate was determined by HPLC, elemental analysis, and DSC FTIR spectroscopy. Sublimation followed a zero-order mechanism and the activation energy and pre-exponential factor were calculated.
The sublimation and vaporisation enthalpies were evaluated and used to determine the melting enthalpy, which was seen to be consistent with DSC data. The solubility parameter, which can be used to predict the miscibility of a drug in a polymer, was calculated from vaporisation enthalpy and crystallographic data.
Finally, the Langmuir equation was used to construct the diagram of the sublimation process, p = f(T). The data were fitted using the Antoine equation by a least squares method.

L’analyse thermogravimétrique a été utilisée pour étudier les processus de sublimation et de vaporisation du S(-) hydrochlorate d’efaroxan, un antagoniste puissant et hautement sélectif des α-2-adrénorécepteurs. Les paramètres cinétiques de la sublimation ont été étudiés par analyse thermogravimétrique conventionnelle isotherme (TGA) et par TGA modulée (MTGA). La caractérisation physico-chimique du sublimate a été effectuée par HPLC, analyse élémentaire, et spectroscopie DSC-FTIR.
